Effect of 2,3,7,8-tetrachlorodibenzo-p-dioxin on the biliary excretion of indocyanine green in rat.

چکیده
Chlorinated dibenzodioxins have been found as contaminants of various technical chlorinated compounds such as 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), which are widely used in agriculture. The dioxin contaminants may be involved in various pathologic states resulting from exposure to technical chlorinated compounds (1, 2). Liver necrosis has been observed in the animals which were treated with derivatives of chlorophenol (3) or other chlorinated compounds (4), and it was suggested that liver necrosis-causing factors were present in these compounds. In view of the above observation, the present study was undertaken to investigate whether 2,3,7,8-tetrachloro-pdibenzodioxin (TCDD) has any effect on the hepatobiliary function of rat. Biliary excretion of indocyanine green (ICG) was used as the index of function. The dye was chosen because it is completely and rapidly excreted by normal liver into the bile by an active process (5).

Differentiation-dependent induction of CYP1A1 in cultured rat small intestinal epithelial cells, colonocytes, and human colon carcinoma cells: basement membrane-mediated apoptosis.
Rat small intestinal epithelial cells and human colon adenocarcinoma cells cultured on Matrigel expressed the differentiation specific enzyme, sucrase-isomaltase, as determined by indirect immunofluorescence. Rat small intestinal epithelial cells, rat colonocytes, and human colon adenocarcinoma cells developed an altered morphology when cultured on Matrigel and became apoptotic within 24-48 h. ...

Unknown, but the effects seem limited. Rats homozygous for the mutant allele have a dramatically increased LD50 for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and PCDD (penta-), but not HCDD (hexa-). Overall, the tissue distribution and excretion of TCDD in rats homozygous for the AhR mutation is similar to that of the Long-Evans rat, which was used as a dioxin-sensitive control strain. Reprodu...
متن کاملExpression of rat sperm flagellum-movement associated protein genes under 2,3,7,8-tetrachlorodibenzo-p-dioxin treatment.
We analyzed the gene expression of sperm flagellum-movement associated proteins, namely, adenylate kinase domain containing protein (RGD1303144) and outer dense fiber protein 1. These gene expressions were up-regulated in a maturing rat testis, and RGD1303144 gene was expressed dominantly in spermatocytes. Although 2,3,7,8-tetrachlorodibenzo-p-dioxin administration reduced sperm motility, these...
متن کاملEffects of ATP depletion with DL-ethionine on biliary excretion of indocyanine green in the rat.
To clarify the energy-dependency of biliary excretion of a diagnostic dye, the effect of ATP depletion on biliary excretion was investigated. Biliary excretion of indocyanine green (ICG) in rat liver reached a maximum 20 min after intravenous application. The level of ATP in the liver was lowered to 35.4% of the control value about 5 h after administration of DL-ethionine (100 mg/100 g body wei...
متن کاملExcretion and tissue distribution of 2,3,7,8-tetrachlorodibenzo-p-dioxin in the rat.
The compound, 2,3,7,8-tetrachlorodibenzop-dioxin (TCDD), is highly toxic. The LD50 for male and female rats given a single oral dose is 23 and 45 jug/kg, respectively (1). Adverse effects have been observed in a teratology study in which pregnant rats were given oral doses of 0.125-2.0 jug/kg-day TCDD from day 6 through day 15 of gestation (2). The adverse effects were increased fetal mortality...
